The Phase 3 SENTRY trial showed that combining selinexor (NEXPOVIO®) with ruxolitinib significantly improved spleen volume reduction by 49.8% at week 24 compared to 28% with ruxolitinib alone in myelofibrosis patients. This combination also showed a promising early overall survival benefit, reducing death risk by over 50%. While symptom improvement was similar to ruxolitinib alone, the spleen reduction was rapid, deep, and sustained, indicating potential disease modification. The treatment was generally well tolerated with manageable side effects. These results, presented at the EHA 2026 Congress, suggest a new therapeutic option for this rare blood cancer with limited current treatments.
