Eli Lilly announced that its Phase 3 BRUIN CLL-322 trial showed adding Jaypirca (pirtobrutinib) to venetoclax and rituximab significantly improved progression-free survival in patients with relapsed or refractory chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL). The combination reduced the risk of disease progression or death by 45% compared to venetoclax and rituximab alone, with median progression-free survival not reached in the Jaypirca arm versus 39.7 months in the control arm. Benefits were seen across high-risk patient groups, and the company plans to submit these results to global regulators to expand Jaypirca’s approved use. Further Phase 3 studies are ongoing to evaluate the therapy in CLL/SLL.
