Cogent Biosciences announced that the PEAK Phase 3 trial showed the combination of bezuclastinib and sunitinib significantly improved progression-free survival (PFS) to 16.5 months compared to 9.2 months with sunitinib alone in gastrointestinal stromal tumor (GIST) patients resistant to imatinib. The combination was effective across all KIT mutation subgroups and had a manageable safety profile similar to sunitinib alone. The FDA granted Priority Review for this combination, with a PDUFA date set for November 30, 2026. Cogent also started a new trial for first-line GIST patients with exon 9 mutations, aiming to expand treatment options.
