Adagene Inc. and Incyte have announced a clinical collaboration to evaluate the combination of muzastotug (ADG126) and INCA33890, a TGFβR2 × PD-1 bispecific antibody, in patients with microsatellite stable colorectal cancer (MSS CRC), including those with liver metastases. The Phase 1 study, sponsored by Incyte and starting in 2026, aims to assess safety, tolerability, and efficacy in chemotherapy-refractory MSS CRC patients. This collaboration builds on promising results from previous trials where muzastotug combined with pembrolizumab showed encouraging response rates. The study could validate muzastotug as a backbone therapy in next-generation immuno-oncology treatments, potentially improving outcomes for a cancer type typically resistant to current immunotherapies.
